Topic:

Kromasil materials have been used in a variety of applications for industrial scale purification of low molecular weight pharmaceuticals, peptides and larger molecules for 30 years today. Key benefits of using Kromasil is its mechanical and chemical stability, as well as its availability in a wide range of particle sizes, so methods can be developed in the early stages of discovery. Reversed phase silica stationary phases such as C18, C8 and C4 is commonly used for the polishing step in a variety of applications for industrial scale purification of low molecular weight pharmaceuticals, peptides and larger molecules for several decades.

In the presentation several examples of method development for large scale HPLC will be shown for peptides, insulins and oligonucleotides. By using crudes with low purity, purification methods were developed by exploring the influence of parameters that needs consideration in large scale purification. These parameters could be stationary phases, organic modifiers, buffers and pH to achieve the most favorable adsorption isotherm. Physical properties of the stationary phases by using elevated temperature and highly alkaline buffers will be illustrated by analytical and preparative injections.

Reversed phase HPCL, is a frequently used technique in the modern pharmaceutical industry from purely analytical usage, to drug discovery and preparative purifications of synthetic molecules, peptides and small proteins. The stationary phases used in these applications are often silica particles coated with C18, C8, or C4 resins, which mainly utilizes hydrophobic interactions, and the optimization tools to could seem limited. However, with some changes of the run conditions, selectivity can increase (or decrease), base line separation between substances can increase, a changed isotherm could significantly improve the purification procedure, large increases in productivity, etc.

This presentation will show how analytical HPLC differs from preparative HPLC, which parameters are important in the two different chromatographic ways; which are not always the same, how to get stable and reproducible chromatography, how and why problems occur, and how to avoid them. Also shown is how purity and yield are increased for a purification of APIs.
